Abstract Phosphonates are organophosphorus compounds possessing a characteristic C−P bond in which phosphorus is directly bonded to carbon. As phosphonates mimic the phosphates and carboxylates of biological molecules potentially inhibit metabolic enzymes, they could be lead for development variety drugs. Fosfomycin (FM) representative phosphonate natural product that widely used as an antibacterial drug. Here, we review biosynthesis FM, includes recent breakthrough find missing link biosynthetic pathway had been mystery quarter-century. In addition, describe genome mining products using gene encoding enzyme catalyzes C–P formation. We also introduce chemoenzymatic synthesis derivatives. These studies expand repertoires related machinery. This mainly covers years 2012-2020.
